         <\/div>\n    <\/div>\n  <\/div>\n<\/section>\n\n\n\n\n\n<p><strong>\u00c9tude de cas du RNRE par&nbsp;<a href=\"https:\/\/www.acorn-nl.ca\/\" target=\"_blank\" rel=\"noreferrer noopener\">ACORN-NL<\/a><\/strong><\/p>\n\n\n\n<p>L\u2019action des m\u00e9dicaments au niveau mol\u00e9culaire ressemble \u00e0 une bo\u00eete noire&nbsp;: habituellement, on se fie sur les r\u00e9sultats des exp\u00e9riences pour tenter de comprendre comment le m\u00e9dicament agit, au lieu de le d\u00e9couvrir physiquement. On sait, par exemple, que les substances contenant du lithium att\u00e9nuent les sympt\u00f4mes de la d\u00e9pression bipolaire, sans vraiment saisir le m\u00e9canisme par lequel elle viennent en aide au cerveau. Cons\u00e9quence&nbsp;: les scientifiques doivent effectuer une foule d\u2019exp\u00e9riences pour r\u00e9unir les pi\u00e8ces du casse-t\u00eate et brosser un tableau des interactions du m\u00e9dicament avec le patient.<\/p>\n\n\n\n<hr class=\"wp-block-separator has-css-opacity\"\/>\n\n\n\n<p>Pourquoi n\u2019arrive-t-on pas \u00e0 \u00e9lucider le fonctionnement d\u2019un m\u00e9dicament au niveau mol\u00e9culaire? Il y a plusieurs raisons \u00e0 cela. Par exemple, il est impossible d\u2019\u00e9tudier de tels processus chez le malade. D\u2019autre part, il est incroyablement difficile d\u2019observer le fonctionnement des mol\u00e9cules pharmaceutiques \u00e0 l\u2019\u00e9chelle sous-microscopique. Enfin les interactions chimiques se produisent \u00e0 une vitesse extr\u00eamement grande. Parce que le tableau des interactions m\u00e9dicamenteuses est incomplet, la d\u00e9couverte d\u2019un nouveau m\u00e9dicament devient donc tr\u00e8s laborieuse, s\u2019accompagne d\u2019effets secondaires malheureux qui n\u2019apparaissent que lors des essais et certaines exp\u00e9riences, en d\u00e9pit de leur utilit\u00e9, deviennent irr\u00e9alisables.<\/p>\n\n\n\n<blockquote class=\"wp-block-quote small is-style-default is-layout-flow wp-block-quote-is-layout-flow\">\n<p><strong><strong>Qu\u2019est-ce que le RNRE?<\/strong><\/strong><\/p>\n\n\n\n<p><em><em>Le R\u00e9seau national de recherche et d\u2019\u00e9ducation (RNRE) est un regroupement d\u2019infrastructures, d\u2019outils et de personnes d\u2019une importance capitale dont la raison d\u2019\u00eatre est de rehausser le leadership du Canada dans les domaines de la recherche, de l\u2019enseignement et de l\u2019innovation. Les partenaires du RNRE canadien coop\u00e8rent afin de rendre cette infrastructure indispensable encore plus s\u00fbre. Un projet pancanadien sans pr\u00e9c\u00e9dent dans l\u2019histoire est en cours pour assurer une surveillance coordonn\u00e9e des menaces qui p\u00e8sent sur le r\u00e9seau.<\/em><\/em><\/p>\n\n\n\n<p><\/p>\n<\/blockquote>\n\n\n\n<h3 class=\"wp-block-heading\">M\u00e9dicaments num\u00e9riques<\/h3>\n\n\n\n<p>Tout cela explique pourquoi les sp\u00e9cialistes en chimie biophysique computationnelle de l\u2019Universit\u00e9 Memorial \u00e0 Terre-Neuve ont con\u00e7u un mod\u00e8le informatique des syst\u00e8mes biologiques simulant l\u2019activit\u00e9 des m\u00e9dicaments \u00e0 l\u2019\u00e9chelle mol\u00e9culaire. En num\u00e9risant les interactions des mol\u00e9cules, les chercheurs reproduisent ce qu\u2019ils ne peuvent observer dans la r\u00e9alit\u00e9. Ils reconstituent n\u2019importe quel processus biologique comme l\u2019absorption des m\u00e9dicaments par la membrane cytoplasmique et d\u2019autres membranes, et cr\u00e9ent image par image une visualisation de ces processus, peu importe la rapidit\u00e9 \u00e0 laquelle ils surviennent ou leur complexit\u00e9.<\/p>\n\n\n\n<p>Comme on pourrait s\u2019y attendre, effectuer et conserver chaque simulation reproduisant avec pr\u00e9cision des millions de mol\u00e9cules qui bougent extr\u00eamement vite \u00e0 l\u2019\u00e9chelle de l\u2019atome exige d\u2019importantes ressources informatiques. Dans cette optique, les chercheurs de l\u2019Universit\u00e9 Memorial exploitent celles d\u2019ACORN-NL, partenaire terre-neuvien du R\u00e9seau national de la recherche et de l\u2019\u00e9ducation (RNRE), qui, avec d\u2019autres partenaires, permet aux \u00e9tablissements d\u2019enseignement sup\u00e9rieur canadien d\u2019acc\u00e9der \u00e0 un r\u00e9seau ultra rapide. Gr\u00e2ce \u00e0 ACORN-NL, les scientifiques de l\u2019Universit\u00e9 Memorial ex\u00e9cutent leurs simulations \u00e0 distance sur le nuage informatique de Calcul Canada et tirent parti d\u2019appareils beaucoup plus performants, dont ils adaptent la puissance en fonction de leurs besoins. On utilise aussi le r\u00e9seau ACORN-NL pour l\u2019analyse, car il autorise le transfert des jeux massifs de donn\u00e9es correspondant aux simulations et aux films virtuels des interactions atomiques entre l\u2019ordinateur des chercheurs et le nuage.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\">La puissance du laboratoire virtuel<\/h3>\n\n\n\n<p>Les simulations aident les scientifiques \u00e0 d\u00e9terminer les particularit\u00e9s structurales de la mol\u00e9cule du m\u00e9dicament qui expliquent son activit\u00e9 dans le corps humain. On s\u2019en servira pour mod\u00e9liser la mani\u00e8re dont les m\u00e9dicaments interagissent dans l\u2019organisme, de leur passage dans l\u2019estomac \u00e0 la travers\u00e9e de la membrane des cellules pour gagner leur cible. Les simulations facilitent et acc\u00e9l\u00e8rent notablement le tri de millions de compos\u00e9s en vue d\u2019isoler ceux qui pourraient se transformer en m\u00e9dicament dans des situations pr\u00e9cises. Les chercheurs peuvent donc se concentrer sur la poign\u00e9e de substances tr\u00e8s prometteuses rep\u00e9r\u00e9es durant la simulation, au lieu de les tester toutes, une \u00e0 une, avec les co\u00fbts exorbitants qu\u2019on imagine. L\u2019ordinateur, \u00e0 qui rien n\u2019\u00e9chappe, peut aussi identifier de fa\u00e7on fiable les ph\u00e9nom\u00e8nes extr\u00eamement rares lors de la simulation, donc rep\u00e9rer les \u00e9v\u00e9nements sporadiques, mais capitaux pour l\u2019\u00e9tude. Ensuite, les chercheurs \u00e9laborent de nouveaux algorithmes pour orienter la simulation sur ces ph\u00e9nom\u00e8nes qui, en d\u00e9pit de leur raret\u00e9, ont souvent de s\u00e9rieuses cons\u00e9quences.<\/p>\n\n\n\n<p>Les possibilit\u00e9s qu\u2019offrent les simulations aident les chercheurs \u00e0 forger de nouveaux m\u00e9dicaments, plus efficaces ou plus faciles \u00e0 fabriquer. Ils peuvent aussi pr\u00e9dire le comportement des m\u00e9dicaments existants lors de nouvelles applications ou modifier ces derniers pour en att\u00e9nuer les effets secondaires ind\u00e9sirables \u2013 autant de r\u00e9alisations qui rehausseront non seulement la qualit\u00e9 de vie des Canadiens ordinaires, mais aussi le bien-\u00eatre de l\u2019humanit\u00e9, en g\u00e9n\u00e9ral. Les scientifiques du monde entier qui cherchent de meilleurs m\u00e9dicaments pour mettre fin \u00e0 la maladie comptent sur l\u2019aide des contributions canadiennes en biophysique computationnelle.
